It is better for criteria cells when they are at their optimal growth rate. The slow freezing process (-1 ° C per minute) also helps to prevent the formation of intracellular ice crystals allowing a sufficient flow of water before freezing. Rapid thawing of cells has the additional purpose of preventing the crystals that have formed during the freezing process from damaging the cell membrane or its components. cryopreservation. DMSO has the drawback of being cytotoxic and therefore cells must be thawed at a faster rate than is possible at room temperature to facilitate the rapid dilution and removal of DMSO from the surrounding cell environment. Cells frozen according to the protocols can be stored for several years in liquid nitrogen. Ideally, frozen cells should not be stored at -80 ºC for long periods of time (up to one week) and should be transferred to liquid nitrogen whenever possible. Seeding cells at -80ºC for prolonged periods may compromise cell viability.
