The blood-brain barrier (BBB) is the interface between the central nervous system (CNS) and the peripheral nervous system (PNS). It is a component of the neurovascular unit (NVU), which prevents circulation of pathogens or toxins that cause brain infections, while allowing essential nutrients to reach the brain. BBBs are not just physical barriers (because of specific tight junctions that prevent unregulated leakage) but they also act as specific transport interfaces (due to specific transporters present on abluminal and luminal membranes) and a metabolic barrier (releasing enzymes locally). Damage or weakening of the BBB allows immune cells to cross it. These cells then attack the myelin around nerves, causing nerve damage and MS symptoms.
